Instructions for the
preparation and use of culture media
- Media must be prepared from dehydrated
culture media, following the instructions stated on
the label.
- Only chemically clean glassware should
be used in dissolving the media.
- It is necessary to weigh accurately
the dehydrated media and to measure exact volumes of
distilled water.
- Dissolve the amount of powder stated
on the label, or small or large quantities, which may
be easily calculated in the desired amount of freshly
distilled or deionized water.
- Heat gently to the boil with frequent
agitation, until complete dissolution of the powder
and sterilize according to the instructions.
- Excessive heating should be avoided.
- It is essential that the pH after
autoclaving be checked regularly. It is especially
important to check the final pH of media prepared from
dehydrated material, which have been opened to detect
changes which may have occurred.

Dehydrated culture
media
1. Materials used in the preparation of
culture media should be dated when received. Care should
be taken to select the oldest batch. Variations may
occur between lots or batches of ingredients. It may be
necessary to adjust media formulations to meet
performance criteria. Complete records should be kept
for each batch.
2. Keep the dehydrated media, in a fresh
and dry place preferably at temperature below 30°C and
at the dark.
3. Check the performance of the prepared
media. Performance criteria are available for many
media.
4. Check the pH of the prepared media.
The pH should be in a range of ± 0.2 of the value
indicated on the label.
5. Avoid keeping bottles opened to long,
when weighing the powder.
6. Keep the bottles gently closed, since
many products are very hygroscopic.
7. Prepared media should be kept at
refrigerator temperature, Plated media should be covered
with aluminium paper or introduced into plastic
bags.

Product storage
and handling
Basic products and ingredients
for culture media
Keep the flask at temperature bellow
30°C, protected from light, in a dry place and perfectly
closed.

Prepared culture media
All rubber capped aluminium tipped
prepared media, may be kept at room temperature with the
same precautions as with dehydrated media. Screw capped
bottles or tubes containing prepared media, should be
manteined at room temperature or at 2-8°C depending on
the product. Follow labelled instructions.

Antibiotic disk
Antibiotic disk containing beta lactam
antimicrobials such as penicillin, oxacillin,
ampicillin, imipenem, piperacillin, and cephalosporins,
shoud be manteined at
-20°C. All other antibiotic disks may be
kept between -20°C and 4°C. Allow the disk to reach room
temperature before use.

Keep at 2-8°C.

Dye reagents
Keep away from light.

Reagents for
identification procedures
Keep at room temperature.

Reagents for immunological or
coagulation test
Keep at 2-8°C except Agarose and V.D.R.L.
which can be manteined at room temperature.
